Design of nucleic acid probe and signal amplification

 

Abstract:

Nucleic acid probe is a fragment of DNA or RNA of variable length which can be designed, labeled, synthesized, and employed at molecular level. It can be used to detect the presence of nucleotide sequences (the DNA or RNA target) that are complementary to the sequence in the probe. Nucleic acids probes have been constructed and utilized in many fields, such as gene analysis, disease diagnosis, protein (enzyme) detection and species taxon.

Here, we present our work on developing nucleic acid probe systems for the detection of miRNA and DNA genome based on signal amplification. It is first presented a highly sensitive, specific and cost-effective strategy to quantify miRNA expression based on two-step RT-qPCR with SYBR-green detection chemistry (we call it as Split-stem RT-qPCR). A target-specific linear primer with split stem is used to realize the aim by circulating the reverse transcription (RT) and qPCR. For the detection of genome, an improved performance of isothermal amplification mediated by a pair of hairpin primers is established, formed only addition a stem-loop structure to a common PCR primer. Similar to traditional PCR, it only employed a DNA polymerase and a pair of primers to realize a three-step cycle process for detection of nucleic acid, but the entire reaction was performed at a single temperature.

 

Biography:

Dr. Xiang Meng received the PhD, MSc and BSc degrees in Analytical Chemistry from Hunan University (2007) and in Microbiology from Hunan normal University in China (2001 and 1993) respectively. After MSc graduation, Meng had worked as a Research Fellow in college of biology, hunan university (2001-), leading a goup working on detection for nucleic acid based on nucleic acid probe for biological/chemical applications, especially in the field of biomedical science.
